Acetyl Hexapeptide-8 is a synthetic hexapeptide produced by solid-phase peptide synthesis (SPPS — solid-phase peptide synthesis). This technology enables precise assembly of the peptide chain from individual amino acids on a solid support, thereby guaranteeing a defined sequence and high product purity. After completion of the synthesis, N-terminal acetylation is carried out, which stabilises the molecule against enzymatic degradation and improves its percutaneous penetration.
The peptide is a structural mimic of the N-terminal segment of the protein SNAP-25 (synaptosomal-associated protein 25) — a key component of the SNARE complex, which controls the release of acetylcholine at the neuromuscular junction. By competitively binding to the SNAP-25 site, Acetyl Hexapeptide-8 limits formation of the SNARE complex, reduces the intensity of muscular microcontractions and decreases the formation and deepening of dynamic expression lines.
SpecPed® SC-AH8 is a pre-formulated aqueous solution from the manufacturer Spec-Chem Industry Inc. (Nanjing, China) with a certified peptide concentration of ≥ 0,05 %. The carrier — a combination of Caprylyl Glycol and Ethylhexylglycerin — simultaneously provides preservative and hydrating functions, which enables direct inclusion in the formulation without further treatment.

Use in formulation

Dosage: 0.5–3 % for everyday use; 3–10 % for intensive care.
Phase: aqueous — disperses homogeneously in both cold and warm water phase.
Solubility: in water.
pH of solution: 5.0–7.0 (according to CoA). Maintain the formulation within this range for optimal peptide stability.
It is recommended to add during the cooling phase (at a temperature below 40 °C) in order to minimise thermal degradation of the peptide bond.

Frequently asked questions

Is Acetyl Hexapeptide-8 in the INCI the same molecule as Acetyl Hexapeptide-3?

Yes, it is the same substance. Acetyl Hexapeptide-3 is the older INCI name, which was updated in 2011 to Acetyl Hexapeptide-8 following revision of the INCI nomenclature. Argireline® is a registered trademark of the original manufacturer Lipotec (BASF). SpecPed® SC-AH8 from Spec-Chem Industry Inc. contains the same active molecule under the generic INCI name.


When using SpecPed® SC-AH8, do I need to add an additional preservative to the formula?

Yes. Caprylyl Glycol and Ethylhexylglycerin are present in the product as carrier excipients (a total of 0.5 % in the raw material). At a dosage of 1–3 % SpecPed SC-AH8 in the final formulation, their resulting concentration is too low to ensure preservation of the product. Aqueous cosmetic preparations always require their own validated preservation system.


Why is the dosage given as a percentage of the entire solution and not of the pure peptide?

SpecPed® SC-AH8 is a pre-formulated solution with a certified peptide concentration ≥ 0.05 %. The dosage refers to the entire solution — it allows direct, accurate measuring without the need to work with concentrated peptide powder or perform your own dilution. The manufacturer guarantees a defined active content (CoA: 0.053 % measured batch 26012601) in every batch.


Can I combine SpecPed® SC-AH8 with vitamin C (ascorbic acid)?

The combination is common but requires attention to pH. Pure ascorbic acid (L-AA) significantly lowers the pH of the formulation (below 3.5), which is outside the recommended range for SpecPed SC-AH8 (5.0–7.0) and may destabilise the peptide. For a stable combination, give preference to more neutral vitamin C derivatives — Ascorbyl Glucoside or Sodium Ascorbyl Phosphate, where the formulation pH remains within a compatible range.


At which stage of manufacture should I add SpecPed® SC-AH8 — before or after emulsification?

After emulsification and cooling of the emulsion below 40 °C. Peptide bonds are sensitive to hydrolysis accelerated by heat, so exposure to emulsification temperature (70–80 °C) is undesirable. In serum formulations without heating (cold water phase), it can be added directly at the beginning — in this case, thermal load is not an issue.


Do I have to store an opened stock in the refrigerator at 2–8 °C?

The requirement of 2–8 °C applies to long-term storage (months) in order to maintain the certified peptide content ≥ 0.05 %. For routine work, a dry, cool and light-protected place is sufficient. It is reasonable to return the opened stock to the refrigerator after each use and to use it up within an appropriate timeframe.
